چکیده

Secretory vesic1es from adrenal medulla contain catecholamines, nuc1eotides and proteins, all of which are released into the extracellular fluid during exocytosis. Adrenal medullary secretory vesic1es also contain high concentrations of Ca'+ [1]. The mechanism of the aecumulation of Ca 2+ into the vesicles is largely unknown and the experimental data eoncerning the uptake of Ca'+ into isolated secretory vesicles are contradictory. It has been reported that secretory vesicle membranes are impermeable to Ca'+ [2], that secretory vesicles take up Ca 2+ independently of ATP [3] and that they possess an ATP-stimulated uptake system [4,5]. In earlier work relatively impure and unstable seeretory vesicle fractions were used for the determination of Ca 2+ -uptake. We have developed a method to isolate highly purified and stable secretory vesicles from bovine medulla [6]. With these vesic1es we repeated earlier Ca'+ -uptake experiments and found that:

Uptake of Ca2+ by isolated secretory vesicles from adrenal medulla.

Intact secretory vesicles isolated from bovine adrenal medulla contain 94 nmol Na+ per mg of protein, and Ca2+ influx into the vesicles is inhibited by increasing concentrations of extravesicular Na+ (but not of K+, Li+ or choline+) or by addition of the Na+ ionophore monensin. Thus Ca2+ influx in determined by the Na+ gradient across the vesicular membrane. Half maximal inhibition of Ca2+ infl...

Inositol 1,3,4,5-tetrakisphosphate-induced Ca2+ sequestration into bovine adrenal-medullary secretory vesicles.

Ins(1,3,4,5)P4 induced a rapid sequestration of Ca2+ into both secretory vesicles and microsomes of bovine adrenal medulla. The Ca(2+)-sequestering role of Ins(1,3,4,5)P4 contrasts with the Ca(2+)-releasing role of Ins(1,4,5)P3 in adrenal-medullary secretory vesicles and microsomes. The Ins(1,3,4,5)P4-induced Ca2+ sequestration into secretory vesicles was not inhibited by heparin (50 micrograms...

Polyphenols of Rubus coreanum Inhibit Catecholamine Secretion from the Perfused Adrenal Medulla of SHRs.

The present study was attempted to investigate whether polyphenolic compounds isolated from wine, which is brewed from Rubus coreanum Miquel (PCRC), may affect the release of catecholamines (CA) from the isolated perfused adrenal medulla of the spontaneously hypertensive rats (SHRs), and to establish its mechanism of action. PCRC (20~180 microg/ml) perfused into an adrenal vein for 90 min relat...

Provinol inhibits catecholamine secretion from the rat adrenal medulla.

The aim of the present study was to examine the effect of provinol, which is a mixture of polyphenolic compounds from red wine, on the secretion of catecholamines (CA) from isolated perfused rat adrenal medulla, and to elucidate its mechanism of action. Provinol (0.3~3 microg/ml) perfused into an adrenal vein for 90 min dose- and time-dependently inhibited the CA secretory responses evoked by A...

Inhibitory effects of olmesartan on catecholamine secretion from the perfused rat adrenal medulla.

The present sutdy aimed to determine whether olmesartan, an angiotensin II (Ang II) type 1 (AT(1)) receptor blocker, can influence the CA release from the isolated perfused model of the rat adrenal medulla. Olmesartan (5~50 µM) perfused into an adrenal vein for 90 min produced dose- and time-dependent inhibition of the CA secretory responses evoked by ACh (5.32 mM), high K(+) (56 mM, a direct m...
